neck alignment / nut alignment

hi i have a jem 7wh, the high e string is a littler closer to the fret edge than the low E, but its not like its gonna fall off or anything. when loosening neck screws, is it ok to have the strings in tune while adjusting? also when retightening neck screws does the order matter?
